Rabbit is the host of choice for producing polyclonal antibodies, and for good reason. As rabbits have been routinely used for a long time, the entire apparatus from rabbit breeds, facilities, to immunisation protocols and secondary antibodies, have been thoroughly tested and evaluated for best performance. There are merits to using chicken, but unless antibody amounts or phylogenetic distance between the host and the species of the target protein are of importance, rabbits remain the most commonly asked for host.
The concentration of total IgG in rabbit antiserum is 5-10 mg/ml, and 1-10% of this are the sought after antigen specific antibodies. A typical bleed results in 20 ml serum, and thus, expected to contain 1-20 mg of specific antibody. The typical yield when purifying a 20 ml serum sample is 1-5 mg.
